Any camera decision will naturally be influenced heavily by the price. The listed launch prices provide an indication of the market segment that the manufacturer of the cameras have been targeting. The NX1 was launched at a markedly lower price (by 85 percent) than the 645D, which puts it into a different market segment. Normally, street prices remain initially close to the MSRP, but after a couple of months, the first discounts appear. Later in the product cycle and, in particular, when the replacement model is about to appear, further discounting and stock clearance sales often push the camera price considerably down. Then, after the new model is out, very good deals can frequently be found on the pre-owned market.
Sensor comparison
The size of the imaging sensor is a crucial determinant of image quality. A large sensor will generally have larger individual pixels that offer better low-light sensitivity, provide wider dynamic range, and have richer color-depth than smaller pixels in a sensor of the same technological generation. Furthermore, a large sensor camera will give the photographer more possibilities to use shallow depth-of-field in order to isolate a subject from the background. On the downside, larger sensors tend to be associated with larger, more expensive camera bodies and lenses.
Of the two cameras under consideration, the Pentax 645D features a medium format sensor and the Samsung NX1 an APS-C sensor. The sensor area in the NX1 is 75 percent smaller. As a result of these sensor size differences, the cameras have a format factor of, respectively, 0.79 and 1.5. The sensor in the 645D has a native 4:3 aspect ratio, while the one in the NX1 offers a 3:2 aspect.
With 39.5MP, the 645D offers a higher resolution than the NX1 (28MP), but the 645D nevertheless has larger individual pixels (pixel pitch of 6.06μm versus 3.63μm for the NX1) due to its larger sensor. However, the NX1 is a much more recent model (by 4 years and 6 months) than the 645D, and its sensor will have benefitted from technological advances during this time that further enhance the light gathering capacity of its pixels. Coming back to sensor resolution, it should be mentioned that neither of the two cameras has an anti-alias filter installed, so they are able to capture all the detail the sensor resolves.
The resolution advantage of the Pentax 645D implies greater flexibility for cropping images or the possibility to print larger pictures. The maximum print size of the 645D for good quality output (200 dots per inch) amounts to 36.3 x 27.2 inches or 92.3 x 69.1 cm, for very good quality (250 dpi) 29.1 x 21.8 inches or 73.8 x 55.3 cm, and for excellent quality (300 dpi) 24.2 x 18.1 inches or 61.5 x 46.1 cm. The corresponding values for the Samsung NX1 are 32.4 x 21.6 inches or 82.3 x 54.9 cm for good quality, 25.9 x 17.3 inches or 65.8 x 43.9 cm for very good quality, and 21.6 x 14.4 inches or 54.9 x 36.6 cm for excellent quality prints.
The NX1 has on-sensor phase detect pixels, which results in fast and reliable autofocus acquisition even during live view operation.
The Pentax 645D has a native sensitivity range from ISO 100 to ISO 1600. The corresponding ISO settings for the Samsung NX1 are ISO 100 to ISO 25600, with the possibility to increase the ISO range to 100-51200.
In terms of underlying technology, the 645D is build around a CCD sensor, while the NX1 uses a BSI-CMOS imager. Both cameras use a Bayer filter for capturing RGB colors on a square grid of photosensors. This arrangement is found in most digital cameras.

Many modern cameras are not only capable of taking still images, but also of capturing video footage. The NX1 indeed provides for movie recording, while the 645D does not. The highest resolution format that the NX1 can use is 4K/30p.
Feature comparison
Beyond body and sensor, cameras can and do differ across a range of features. For example, the NX1 has an electronic viewfinder (2360k dots), while the 645D has an optical one. One differentiating feature between the two cameras concerns the touch sensitivity of the rear screen. The NX1 has a touchscreen, while the 645D has a conventional panel. Touch control can be particularly helpful, for example, for setting the focus point.

It is notable that the NX1 offers wifi support, which can be a very convenient means to transfer image data to an off-camera location. In contrast, the 645D does not provide wifi capability.
