Any camera decision will naturally be influenced heavily by the price. The listed launch prices provide an indication of the market segment that the manufacturer of the cameras have been targeting. The A9 was launched at a markedly lower price (by 55 percent) than the 645D, which puts it into a different market segment. Usually, retail prices stay at first close to the launch price, but after several months, discounts become available. Later in the product cycle and, in particular, when the replacement model is about to appear, further discounting and stock clearance sales often push the camera price considerably down.
The size of the sensor inside a digital camera is one of the key determinants of image quality. All other things equal, a large sensor will have larger individual pixel-units that offer better low-light sensitivity, wider dynamic range, and richer color-depth than smaller pixels in a sensor of the same technological generation. Further, a large sensor camera will give the photographer additional creative options when using shallow depth-of-field to isolate a subject from its background. On the downside, larger sensors tend to be more expensive and lead to bigger and heavier cameras and lenses.
Of the two cameras under consideration, the Pentax 645D features a medium format sensor and the Sony A9 a full frame sensor. The sensor area in the A9 is 42 percent smaller. As a result of these sensor size differences, the cameras have a format factor of, respectively, 0.79 and 1.0. The sensor in the 645D has a native 4:3 aspect ratio, while the one in the A9 offers a 3:2 aspect.
With 39.5MP, the 645D offers a higher resolution than the A9 (24MP), but the 645D nevertheless has larger individual pixels (pixel pitch of 6.06μm versus 5.94μm for the A9) due to its larger sensor. However, the A9 is a much more recent model (by 7 years and 1 month) than the 645D, and its sensor will have benefitted from technological advances during this time that further enhance the light gathering capacity of its pixels. Coming back to sensor resolution, it should be mentioned that the 645D has no anti-alias filter installed, so that it can capture all the detail its sensor resolves.
The resolution advantage of the Pentax 645D implies greater flexibility for cropping images or the possibility to print larger pictures. The maximum print size of the 645D for good quality output (200 dots per inch) amounts to 36.3 x 27.2 inch or 92.3 x 69.1 cm, for very good quality (250 dpi) 29.1 x 21.8 inch or 73.8 x 55.3 cm, and for excellent quality (300 dpi) 24.2 x 18.1 inch or 61.5 x 46.1 cm. The corresponding values for the Sony A9 are 30 x 20 inch or 76.2 x 50.8 cm for good quality, 24 x 16 inch or 61 x 40.6 cm for very good quality, and 20 x 13.3 inch or 50.8 x 33.9 cm for excellent quality prints.
The A9 has on-sensor phase detect pixels, which results in fast and reliable autofocus acquisition even during live view operation.
The Pentax 645D has a native sensitivity range from ISO 100 to ISO 1600. The corresponding ISO settings for the Sony Alpha A9 are ISO 100 to ISO 51200, with the possibility to increase the ISO range to 50-204800.
Since 2007, DXO Mark has published sensor performance measurements that have been derived using a consistent methodology. This service is based on lab testing and assigns an overall score to each camera sensor, as well as ratings for dynamic range ("DXO Landscape"), color depth ("DXO Portrait"), and low-light sensitivity ("DXO Sports"). Of the two cameras under consideration, the A9 has a markedly higher DXO score than the 645D (overall score 10 points higher), which will translate into better image quality. Many modern cameras are not only capable of taking still images, but can also record movies. The A9 indeed provides for movie recording, while the 645D does not. The highest resolution format that the A9 can use is 4K/30p.
Apart from body and sensor, cameras can and do differ across a range of features. For example, the A9 has an electronic viewfinder (3686k dots), while the 645D has an optical one. Both systems have their advantages, with the electronic viewfinder making it possible to project supplementary shooting information into the framing view, whereas the optical viewfinder offers lag-free viewing and a very clear framing image. One feature that is present on the 645D, but is missing on the A9 is a top-level LCD. While being, of course, smaller than the rear screen, the control panel conveys some of the essential shooting information and can be convenient for quick and easy settings verification.
The reported shutter speed information refers to the use of the mechanical shutter. Yet, some cameras only have an electronic shutter, while others have an electronic shutter in addition to a mechanical one. In fact, the A9 is one of those camera that have an additional electronic shutter, which makes completely silent shooting possible. However, this mode is less suitable for photographing moving objects (risk of rolling shutter) or shooting under artificial light sources (risk of flickering).
The Pentax 645D has an intervalometer built-in. This enables the photographer to capture time lapse sequences, such as flower blooming, a sunset or moon rise, without purchasing an external camera trigger and related software.
The 645D writes its imaging data to SDXC cards, while the A9 uses SDXC or Memory Stick PRO Duo cards. Both cameras feature dual card slots, which can be very useful in case a memory card fails. The A9 supports UHS-II cards on its first slot and UHS-I on its second one, while the 645D cannot take advantage of Ultra High Speed SD cards.

It is notable that the A9 offers wifi support, which can be a very convenient means to transfer image data to an off-camera location. In contrast, the 645D does not offer wifi capability.
