Any camera decision will obviously take relative prices into account. The listed launch prices provide an indication of the market segment that the manufacturer of the cameras have been targeting. The X-Pro2 was launched at a markedly lower price (by 83 percent) than the 645D, which puts it into a different market segment. Normally, street prices remain initially close to the MSRP, but after a couple of months, the first discounts appear. Later in the product cycle and, in particular, when the replacement model is about to appear, further discounting and stock clearance sales often push the camera price considerably down.
The imaging sensor is at the core of digital cameras and its size is one of the main determining factors of image quality. A large sensor will tend to have larger individual pixels that provide better low-light sensitivity, wider dynamic range, and richer color-depth than smaller pixel-units in a sensor of the same technological generation. Furthermore, a large sensor camera will give the photographer more possibilities to use shallow depth-of-field in order to isolate a subject from the background. On the downside, larger sensors tend to be more expensive and lead to bigger and heavier cameras and lenses.
Of the two cameras under consideration, the Fujifilm X-Pro2 features an APS-C sensor and the Pentax 645D a medium format sensor. The sensor area in the 645D is 295 percent bigger. As a result of these sensor size differences, the cameras have a format factor of, respectively, 1.5 and 0.79. The sensor in the X-Pro2 has a native 3:2 aspect ratio, while the one in the 645D offers a 4:3 aspect.
With 39.5MP, the 645D offers a higher resolution than the X-Pro2 (24MP), but the 645D nevertheless has larger individual pixels (pixel pitch of 6.06μm versus 3.92μm for the X-Pro2) due to its larger sensor. However, the X-Pro2 is a much more recent model (by 5 years and 10 months) than the 645D, and its sensor will have benefitted from technological advances during this time that enhance the light gathering capacity of its pixel-units. Coming back to sensor resolution, it should be mentioned that neither of the two cameras has an anti-alias filter installed, so they are able to capture all the detail the sensor resolves.
The resolution advantage of the Pentax 645D implies greater flexibility for cropping images or the possibility to print larger pictures. The maximum print size of the 645D for good quality output (200 dots per inch) amounts to 36.3 x 27.2 inch or 92.3 x 69.1 cm, for very good quality (250 dpi) 29.1 x 21.8 inch or 73.8 x 55.3 cm, and for excellent quality (300 dpi) 24.2 x 18.1 inch or 61.5 x 46.1 cm. The corresponding values for the Fujifilm X-Pro2 are 30 x 20 inch or 76.2 x 50.8 cm for good quality, 24 x 16 inch or 61 x 40.6 cm for very good quality, and 20 x 13.3 inch or 50.8 x 33.9 cm for excellent quality prints.
The X-Pro2 has on-sensor phase detect pixels, which results in fast and reliable autofocus acquisition even during live view operation.
The Fujifilm X-Pro2 has a native sensitivity range from ISO 200 to ISO 12800, which can be extended to ISO 100-51200. The corresponding ISO settings for the Pentax 645D are ISO 100 to ISO 1600 (no boost).

Many modern cameras cannot only take still pictures, but also record videos. The X-Pro2 indeed provides movie recording capabilities, while the 645D does not. The highest resolution format that the X-Pro2 can use is 1080/60p.

One feature that is present on the 645D, but is missing on the X-Pro2 is a top-level LCD. While being, of course, smaller than the rear screen, the control panel conveys some of the essential shooting information and can be convenient for quick and easy settings verification.
The reported shutter speed information refers to the use of the mechanical shutter. Yet, some cameras only have an electronic shutter, while others have an electronic shutter in addition to a mechanical one. In fact, the X-Pro2 is one of those camera that have an additional electronic shutter, which makes completely silent shooting possible. However, this mode is less suitable for photographing moving objects (risk of rolling shutter) or shooting under artificial light sources (risk of flickering).
The Fujifilm X-Pro2 and the Pentax 645D both have an intervalometer built-in. This enables the photographer to capture time lapse sequences, such as flower blooming, a sunset or moon rise, without purchasing an external camera trigger and related software.
Concerning the storage of imaging data, both the X-Pro2 and the 645D write their files to SDXC cards. Both cameras feature dual card slots, which can be very useful in case a memory card fails. The X-Pro2 supports UHS-II cards on its first slot and UHS-I on its second one, while the 645D cannot take advantage of Ultra High Speed SD cards.
